IFNa2b-IFNa5, IFNa5-IFNa2b, IFNa5-IFNa5 chimerinių genų klonavimas ir raiška E.coli BL21 (DE3) ląstelėse
Abstract
Baigiamajame magistro darbe aprašomas chimerinių IFNα2b-IFNα5, IFNα5-IFNα2b ir IFNα5-IFNα5 genų konstravimas, klonavimas ir raiška E. coli BL21 (DE3) ląstelėse atliekant auginimus kolbose. Išbandyti ir palyginti trys alternatyvūs IFNα5-IFNα5 ir penki IFNα2b-IFNα5 geno konstravimo metodai, pasirinkti efektyviausi variantai, o IFNα5-IFNα2b konstruotas analogiškai IFNα2b-IFNα5. Chimeriniai genai klonuoti į E. coli BL21 (DE3) ląsteles naudojant raiškos vektorių pET21a+. Visų genetinių konstrukcijų tapatumas patikrintas restrikcine analize bei DNR sekoskaita. Atliktos tikslinių baltymų raiškos E.coli BL21 (DE3) ląstelėse analizės, įsitikinta, kad po indukcijos IPTG chimeriniai baltymai efektyviai sintetinami ir ląstelėse aptinkami netirpios formos. Parinktos sukonstruotų producentų auginimo kolbose sąlygos. Sukonstruoti rekombinantiniai baltymai gali būti naudojami tirti IFNα5 įtaką hepatito C bei kitų virusinių infekcijų ir vėžinių susirgimų, kurių atvejais skiriami interferoniniai preparatai, gydymui, kai kartu į organizmą patenka toks pat kiekis IFNα2b (išskyrus IFNα5-IFNα5), papildomiems baltymo-receptoriaus sąveikos tyrimams ir tiriant tirpios formos serume aptinkamus receptorius, slopinančius interferono poveikį. Galima būtų nustatyti, ar galima mažinti plazmoje aptinkamų tirpių interferonų receptorių įtaką baltymo biologinio poveikio slopinimui naudojant rekombinantinį dimerą, sudarytą iš dviejų viso ilgio interferono molekulių. Darbą sudaro 6 dalys: įvadas, literatūros apžvalga, medžiagos, metodai, aparatūra, rezultatai ir jų aptarimas, išvados, literatūros sąrašas. The aim of this work is to describe construction, cloning and expression of IFNα2b-IFNα5, IFNα5-IFNα2b and IFNα5-IFNα5 fusion genes in E. coli BL21 (DE3) cells. Three alternative strategies were tested for constructing IFNα5-IFNα5 gene and five for IFNα2b-IFNα5 gene, the most effective strategies were chosen. IFNα5-IFNα2b construction scheme is analogous to IFNα2b-IFNα5. Fusion genes were cloned into E. coli BL21 (DE3) cells using pET21a+ expression vector. Restriction analysis and DNA sequencing were used to confirm the identity of the obtained recombinant vectors. Also expression analysis of target proteins was performed, the results show that the expression is effective after IPTG induction and these proteins are insoluble in bacterial cells. Afterwards the growth conditions for recombinant bacteria were chosen. Created recombinant proteins could be used to further explore the influence of IFNα5 in treatment of hepatitis C and other viral infections and cancers, when the same amount of IFNα2b is used together (except IFNα5-IFNα5). Also these proteins could be helpful in analyzing protein-receptor interactions between interferon and its receptor, including the soluble receptors found in serum. It could be revealed whether it is possible to reduce the impact of soluble interferon receptors on the protein biological activity reduction using the recombinant protein comprised of two full-length interferon molecules.
