Gliukozės biologinio jutiklio kūrimas biologiškai aktyvių junginių bei cistamino dihidrochlorido pagrindu.
Abstract
Sergamumas cukriniu diabetu pasaulyje sparčiai didėja, todėl mokslininkų dėmesys skiriamas gliukozės biologinių jutiklių konstrukcijos tyrimams. Pastaraisiais metais biologinių jutiklių kūrimo technologijos sparčiai vystėsi, padaryta pažanga kuriant nanodydžio BJ, kuriuose naudojamos įvairios nanomedžiagos. Darbo tikslas – sukurti jautrų bei stabilų laike gliukozės biologinį jutiklį biologiškai aktyvių junginių bei cistamino dihidrochlorido pagrindu ir pritaikyti modelinę sistemą praktikoje. Sukurtos 7 elektrodų sistemos ir nustatytos 3 didžiausią analizinį signalą generavusios: GE/CADHCl/AuND/FD/GOx/GA, GE/AuNS/CADHCl/AuND/FD/GOx/GA, GE/AuND/CADHCl/FD/GOx/GA. Nustatyta, kad GE turi būti laikomas 5 mM CADHCl tirpale 16 val. kambario temperatūroje (+21 oC), o elektrocheminiams tyrimams patartina naudoti chronoamperometrinį metodą, esant +0,30 V potencialui. Pastebėta, kad BJ yra tinkami elektrocheminei analizei dėl oksidacijos-redukcijos proceso grįžtamumo. Nustatyta, kad biologinis jutiklis GE/AuND/CADHCl/FD/GOx/GA pagrindu pasižymi geromis analizinėmis charakteristikomis: 0,1–22,9 mM tiesiniu gliukozės koncentracijos intervalu, 0,9969 koreliacijos koeficientu, 6,89 % variacijos koeficientu (esant 7,2 mM Glc) bei 87 % atsikartojamumu (esant 4,48 mM Glc ir 16 matavimų) bei mažiausia aptikimo riba (0,088 mM) ir didžiausiu stabilumu (27 paros), lyginant su GE/CADHCl/AuND/FD/GOx/GA ir GE/AuNS/CADHCl/AuND/FD/GOx/GA. Taip pat GE/AuND/CADHCl/FD/GOx/GA pagrindu sukurtas BJ modelis buvo sėkmingai pritaikomas žmogaus seilėse bei serume, migdolų piene bei mandarinų sultyse, nes pasižymėjo atsparumu trukdžiams ir geromis gliukozės išgavomis žmogaus seilėse: 96,0–98,6 %, žmogaus serume: 86,0–97,1 %, migdolų piene: 90,0–98,7 % ir mandarinų sultyse: 85,0–95,7 %. Diabetes rates are increasing worldwide, prompting extensive research on glucose biosensors. Nanoscale biosensors using various nanomaterials have shown significant advancements in recent years.The aim of this work is to develop a sensitive and time-stable glucose biosensor based on biologically active compounds and cystamine dihydrochloride and to apply the model system in practice. 7 electrode systems were developed and the 3 electrodes with the highest analytical signal were identified: GE/CADHCl/AuNP/PD/GOx/GA, GE/AuNS/CADHCl/AuNP/PD/GOx/GA and GE/AuNP/CADHCl/PD/GOx/GA. It has been established that graphite rod electrode should be stored in 5 mM CADHCl solution for 16 hours at room temperature (+21 oC), and that for electrochemical analysis, the chronoamperometric method at a potential of +0.30 V is recommended. The reversibility of the oxidation-reduction process has been observed to make the biosensors suitable for electrochemical analysis. The GE/AuNP/CADHCl/PD/GOx/GA based biosensor was found to have good analytical performance: 0.1-22.9 mM of linear glucose concentration range, 0.9969 of correlation coefficient, 6.89 % of variation coefficient (at 7.2 mM Glc) and 87 % of reproducibility (at 4,48 mM of Glc and 16 measurements), the lowest limit of detection (0,088 mM) and the highest stability (27 days) compared to GE/CADHCl/AuNP/PD/GOx/GA and GE/AuNS/CADHCl/AuNP/PD/GOx/GA. In addition, the GE/AuNP/CADHCl/PD/GOx/GA based biosensor model has been successfully applied to human saliva and serum, almond milk and mandarin juice, due to its resistance to interference and good glucose recoveries in human saliva solution: 96,0-98,6 %, in human serum solution: 86,0-97,1 %, in almond milk: 90,0-98,7 % and in mandarin juice: 85,0-95,7 %.
