Toxicity of α-synuclein and Aβ40 in Saccharomyces cerevisiae
Abstract
Saccharomyces cerevisiae expression and secretion systems bearing glucose dehydrogenase gene fused with A β40 or SNCA sequences were generated and applied to study toxicity of amyloid- β and α-synuclein in yeast. Analysis of transformant growth demonstrated that cytoplasm-localized A β40 peptide exhibits higher toxicity to the yeast cells than α-synuclein. Arrangement of A β-bearing hybrid to the secretory pathway reduced the toxic effect for host cells.
