Mutavusio huntingtino baltymo agregatai R6/2 pelių tinklainėje ir regos nerve
Abstract
Šio darbo tikslas įvertinti mutavusio huntingtino baltymo įtaką tinklainės ganglinėms ląstelėms ir regos nervui. Tinklainės ir regos nervai buvo išpreparuoti iš skirtingo amžiaus R6/2 (Huntingtono ligos modelis pelėse) ir laukinio tipo pelių. TUNEL metodu buvo nustatyta, kad tinklainės nervinės ląstelės nežūsta, nors jose ir kaupiasi mutavęs huntingtino (mHtt) baltymas. Ligai progresuojant ląstelėse kaupiasi vis daugiau mHtt ir palaipsniui tirpi baltymo forma pereina į agreguotą formą. Imunofluorescenciniu dažymu buvo nustatyta, kad tinklainės astrocitai neturi savo somoje mHtt, o regos nervo astrocitai kaupia mHtt. Taip pat buvo pastebėta, kad optiniame nerve mHtt depozitų skaičius didėja ligai progresuojant. Imunohistochemiškai nudažius ląsteles Lamp2 ir ubikvitinu nebuvo pastebėta jokių ženklių pakitimų, tačiau buvo žymiai padidėjęs p62 baltymo kiekis transgeninėse pelėse Tiek dryžuotame kūne tiek tinklainėje LC3-I, LC3-II ir Hsp70 pokyčiai nebuvo reikšmingi lyginant skirtumus tarp atskirų amžiaus grupių ir tarp laukinio tipo ir transgeninių pelių. Tačiau nors šiuose mechanizmuose yra sutrikimų, tinklainės ganglinės ląstelės nežūsta. Darbo apimtis: 47 p. teksto be priedų, 16 paveikslų, 1 lentelė, 62 bibliografiniai šaltiniai The aim of this work was to examinate the effect of mutant huntingtin (mHtt) protein on retinal ganglion cell survival and optic nerve degeneration. Retinas and optic nerves were isolated from different age R6/2 transgenic mice and their wild type (WT) littermates. TUNEL staining showed that young 4-week-old animals had the lowest number of TUNEL-positive cells in the outer nuclear layer (ONL), whereas the 12-week-old R6/2 mice had the highest number of apoptotic cells in the ONL. However, no TUNEL positive cells were found in the retinal ganglion cell layer (RGCL). The mHtt staining in retinas of R6/2 mice was found at the age of 4 weeks with only a few cells having aggregates that are positive for mHtt. At the age of 8 weeks the number of RGCL cells that had mHtt positive aggregates significantly increased as compared to the 4-week group. At later age groups the majority of RGCL cells contained aggregated form of mHtt. The R6/2 mice did not differ from their WT littermates in the total number of retinal astrocytes or total number of cells in the RGCL at any age. Immunohistochemical stainings of Lamp2, p62, ubiquitin, LC3 and Hsp70 proteins were performed to detect changes in protein degradation pathways. The number of RGCL cells that had p62 - immunoreactivity was significantly increased in the 4-week, 12-week and 16-week R6/2 retinas as compared to WT mice. Expressions of Lamp2 and ubiquitin in the cells were the same in R6/2 and WT mice.
